Title: Effect of Certain Heavy Metals on the Activity of ATPases

Abstract: Heavy metals have received considerable attention over the recent years as a result of increased environmental pollutants from industrial , agricultural, energy and municipal sources . Cadmium, lead, and other heavy metals have been spread to the soil for the last decades by sewage used as fertilizer, or via irrigation or airborne pollution . in vitro, effect of cadmium, lead, and mercury were studied as an energy transfer inhibitors of chloroplasts Mg+2-ATPase activity and Ca+2-ATPase activity of chloroplasts coupling factor .Chloroplasts isolated from spinach ( Spinacia oleraceae ) leaves that irrigated with well water or treated municipal waste water ( TMWW ) . Data showed that cadmium is the one from the three tested heavy metals that affect both membrane bound ATPase and soluble coupling factor 1 . Kinetically cadmium inhibits Mg+2- ATPase and Ca+2-ATPase competitively with Ki values of 15.2  1.8 M and 48.2  5.1 M respectively . On the other hand, lead and mercury had slight effect on the two enzyme systems . For the plants that were irrigated with TMWW, cadmium, lead and mercury stimulate Ca+2-ATPase activity at low concentrations ( up to ~ 50 M ). Cadmium at 100 M inhibits 28 % of Ca+2 - ATPase while, lead and mercury had no effect .
